Besides contamination with yeast, fungi and other cell types, the contamination of cell cultures with viruses, bacteria and mycoplasma is widely spread. Up to 87 % of all cell cultures in academic use and up to 4 % in industrial production are contaminat­ed with mycoplasma. Approx. 6 % of all cells used for therapeutic purposes are contami­nated with undetected bacteria. The regular use of antibiotics in cell culture media suppresses the bacterial growth to a level which are undetectable without a suitable detection method.

Impact
It is important to keep an eye on these con­taminants during routine work out of diversified and severe reasons: Inhibition of cell proliferation up to 50 %, higher infection risk for patients treated with therapeutic cells, unspecific positive results in animal control testing by stimulated immune systems, influence of virus proliferation and infection rates, disturbance of the hybridoma-technique and patch-clamp experiments as well as a drastic decrease of electroporation rates. Mycoplasma can constitute up to 50 % of the total protein and 15 - 30 % of the DNA isolated from a cell culture. These impurities will complicate any downstream purification processes and cloning experi­ments.

Consequences
The PCR method can easily fulfill the restriction of the guideline in means of specificity and robustness. The sensitivity of the test method is limited by the low sample volume. This problem was solved by a cell culture enrichment which also solves another problem of direct testing: the detection of DNA from dead mycoplasma. Spiking a cell culture with 10 vital mycoplasma particles gives a strong detectable PCR signal within one week of cultivation; DNA from dead mycoplasma will be digested by extracellular nucleases.
A PCR method validated according to the guidelines of the Eur. Ph., e.g. Minerva Biolabs´ Venor®GeM-qEP for qPCR, is a powerful and easy to use tool for enhanced mycoplasma safety.
